چکیده

Methods Ninety seven JIA patients with a polyarticular course were enrolled into the study. All patients received methotrexate treatment for at least for 6 months. The efficacy of treatment was judged by improvement in physician’s global assesment, the number of affected joints, erythrocyte sedimentation rate and C-reactive protein. RFC and ATIC gene polymorphisms were determined by polymerase chain reactionrestriction fragment length polymorphism ( PCR-RFLP) analysis. Results The median age of the patients was 72 (12-92) months. The response rate to MTX was 45%. There was no significant association between RFC and ATIC polymorphisms and efficacy of MTX treatment (p: 0.254, p: 0.098 respectively).

Association of the 5-aminoimidazole-4-carboxamide ribonucleotide transformylase gene with response to methotrexate in juvenile idiopathic arthritis

OBJECTIVES Methotrexate (MTX) is the mainstay treatment for juvenile idiopathic arthritis (JIA), however approximately 30% of children will fail to respond to the drug. Identification of genetic predictors of response to MTX would be invaluable in developing optimal treatment strategies for JIA. Using a candidate gene approach, single nucleotide polymorphisms (SNPs) within genes in the metaboli...

Outcomes of methotrexate therapy for psoriasis and relationship to genetic polymorphisms

BACKGROUND The use of methotrexate is limited by interindividual variability in response. Previous studies in patients with either rheumatoid arthritis or psoriasis suggest that genetic variation across the methotrexate metabolic pathway might enable prediction of both efficacy and toxicity of the drug. OBJECTIVES To assess if single nucleotide polymorphisms (SNPs) across four genes that are ...

Inhibition of 5-aminoimidazole-4-carboxamide ribotide transformylase, adenosine deaminase and 5'-adenylate deaminase by polyglutamates of methotrexate and oxidized folates and by 5-aminoimidazole-4-carboxamide riboside and ribotide.

With the use of a continuous spectrophotometric assay and initial rates determined by the method of Waley [Biochem. J. (1981) 193, 1009-1012] methotrexate was found to be a non-competitive inhibitor, with Ki(intercept) = 72 microM and Ki(slope) = 41 microM, of 5-aminoimidazole-4-carboxamide ribotide transformylase, whereas a polyglutamate of methotrexate containing three gamma-linked glutamate ...

Single-nucleotide polymorphisms in the folate pathway are associated with response to methotrexate treatment in juvenile idiopathic arthritis

Methods Genotypes were determined in an observational cohort of 183 JIA patients that had been systematically followed at 3 months intervals. The following SNPs were determined: methylenetetrahydrofolate reductase (MTHFR) 677C>T and 1298A>C, methionine synthase reductase (MTRR) 66A>G, thymidylate synthase (TS) 2R/3R and Reduced Folate Carrier (RFC) 80G>A. MTX efficacy and adverse effects were c...

Crystal structures of human bifunctional enzyme aminoimidazole-4-carboxamide ribonucleotide transformylase/IMP cyclohydrolase in complex with potent sulfonyl-containing antifolates.

Aminoimidazole-4-carboxamide ribonucleotide (AICAR) transformylase/IMP cyclohydrolase (ATIC) is a bifunctional enzyme with folate-dependent AICAR transformylase and IMP cyclohydrolase activities that catalyzes the last two steps of purine biosynthesis. The AICAR transformylase inhibitors BW1540 and BW2315 are sulfamido-bridged 5,8-dideazafolate analogs with remarkably potent K(i) values of 8 an...
